STRAIGHT TO THE SOLUTION

Test the microphone in Windows Sound. Unmute its hardware switch and select the correct input.

Review Dota 2 audio input and push-to-talk key. Make sure you are using the intended voice channel.

Allow desktop microphone access and restart the game after selecting the device.

Must I reinstall the game?

Reinstallation usually does not fix a wrong input or missing permission. Check these settings first.
